Natural phytochemicals such as curcumin with 3-ABN and also their synthetic derivatives have received the attention of various chemist and biochemist owing to their various pharmacological actions. Development and discovery of new 3-ABN based hydrazine derivatives with antioxidant activities is one of the interesting areas in medicinal and natural product chemistry. In the present study, a new series of 3-ABN-based hydrazine derivative was synthesized by the modified and an optimized successive double Mannich condensation method and characterization was carried out by HNMR, CNMR and IR and Elemental analysis methods. In vitro free radical scavenging and anti-microbial activity was evaluated against various free radicals, bacteria’s and fungus strains. All the compounds (8-13) displayed radical scavenging activity in a concentration–dependent manner against various free radicals because of their hydrogen/electron donating functionalities. However, the activity was various with the different substitution at the C-2 and C-6 positions of the azabicyclononan-9-one moiety. Compared to standard antioxidant ascorbic acid, Compounds 8 and 12 showed excellent free radical scavenging effects due to the incorporation of methyl groups at para position phenyl ring. These data present a series of novel synthesized 3-ABN based-hydrazone derivative; especially compounds 8 and 12 could be suggested as potential antioxidant lead compounds. Although we found admirable in vitro free radical scavenging effects against various free radicals in compounds possessing electron-withdrawing chloro (9/13), bromo (10), and fluoro (11), substitutions at the para position of the azabicycloninan-9-one moiety, these compounds exhibited outstanding antibacterial and antifungal activities owing to their pro-oxidant effects. Our findings suggest compounds 9,10,11 has activity against the various microbes (1-56-6.25uM/mL) and could be developed as a potential therapeutic agent.
